Research in Endocrinology, aimed at the investigation of the cellular signaling, gene expression and hormone secretion in anterior pituitary cells. • Routinely performed isolation, optimization and maintenance of rat and mouse pituitary primary cell-cultures and maintained various cancer cell lines in the laboratory; • Performed radioimmunoassays (RIA) and enzyme-linked immunosorbent assays (ELISA) of secreted pituitary hormones and cAMP/cGMP nucleotides from perifused and cultured pituitary cells in normal and pharmacologically challenged conditions; • Extensively analyzed RNA transcriptome and proteome of GnRH-controlled gene network of rat pituitary gonadotrophs using Next Generation Sequencing (RNA-Seq), TaqMan qRT-PCR analysis and Western Blotting; • Recombinantly expressed and purified tagged and fluorescently labeled fusion proteins, validated commercially available antibodies; • Performed and evaluated siRNA efficacy of gene silencing primarily by qRT-PCR and Western Blotting; • Conducted standard laboratory techniques such as DNA/RNA isolation, DNA gel electrophoresis, restriction analysis, plasmid preparation and extraction, standard and dose response curve generation, maintenance of laboratory stocks and preparation of reagents; • Supervised and trained new staff and mentored summer students. Research focused on the identification and the characterization of steroid metabolites and steroid metabolizing enzymes in chicken and rat intestine and other tissues. • Solid-state extraction and HPLC/Mass spectrometry of steroid hormones and their metabolites from various rat and chicken tissues with emphasis on the activity of glucocorticoid metabolizing enzymes (11β-HSD1 and 2); • Identification, bacterial recombinant expression and affinity column purification of a novel chicken 20β-hydroxy-steroid dehydrogenase enzyme (20β-HSD); • Gene expression analysis, RNA extraction, plasmid preparation, transfection and extraction; • Performed enzyme activity assays on native and recombinant steroid-metabolizing enzymes, identified their steroid ligands and metabolites and evaluated the effect of these modifications on steroid receptor binding; • Assisted with writing of grants and publications.
