      I carried out the master's thesis "Selection and characterization of variants of a bacterial formate dehydrogenase obtained by directed evolution" (in English) in the laboratory of Molecular Biology and Biocatalysis of Prof. Elisabetta Bergantino at the Department of Biology of the University of Padua.During the training, I was involved in the expression and characterization of variants of a bacterial formate dehydrogenase previously obtained by directed evolution. In this way, I was able… I carried out the master's thesis "Selection and characterization of variants of a bacterial formate dehydrogenase obtained by directed evolution" (in English) in the laboratory of Molecular Biology and Biocatalysis of Prof. Elisabetta Bergantino at the Department of Biology of the University of Padua.During the training, I was involved in the expression and characterization of variants of a bacterial formate dehydrogenase previously obtained by directed evolution. In this way, I was able to gain experience in the preparation of PCR, SDS-PAGE and Western-Blot reactions, in the cloning of gene inserts in plasmid vectors, in the use of E. coli as an expression system (fromtransformation and selection of the transformed up to cell culture and induction of the expression) and in the expression of recombinant proteins and their purification by affinity column (IMAC).ITALIANOHo svolto la tesi magistrale "Selection and characterization of variants of a bacterial formate dehydrogenase obtained by directed evolution" (in inglese) nel laboratorio di Biologia Molecolare e Biocatalisi della prof.ssa Elisabetta Bergantino presso il Dipartimento di Biologia dell'Università degli studi di Padova.Durante il tirocinio mi sono occupato dell'espressione e caratterizzazione di varianti di una formiato deidrogenasi batterica precedentemente ottenute tramite evoluzione diretta. In questo modo ho avuto modo di acquisire esperienza nell'allestire reazioni di PCR, SDS-PAGE e Western-Blot, nel clonaggio di inserti genici in vettori plasmidici, nell'uso di E. coli come sistema di espressione (dallatrasformazione e selezione dei trasformati fino alla coltura di cellule e all'induzione dell'espressione) e nell'espressione di proteine ricombinanti e loro purificazione mediante colonna di affinità (IMAC).       I carried out the bachelor's thesis "Evaluation of cytotoxic effects of free and encapsulated gemcitabinein nanoparticles on human lymphocytes" in the laboratory of Prof. Susanna Mandruzzato at the Veneto Oncological Institute (IOV) - IRCCS of Padua.During the training, I compared free gemcitabine with that encapsulated in nanoparticles, to determine if the latter was able to decrease the cytotoxic effect of the drug on human lymphocytes. Ho fatto esperienza utilizzando il saggio colorimetrico MTT, coltivando cellule tumorali (fibroblasti) e lavorando in sterilità sotto cappa biologica. Show more Show less
