      Komor Research Group Faculty Advisors: Dr. Alexis Komor & Dr. Francesco Paesani Ph.D. Student Mentors: Carlos Vasquez & Kartik Lakshmi My research is focused on characterizing point mutations in a DNA repair gene that are inherited by patients with an increased risk of developing colorectal cancer. To investigate the effect of individual mutations on DNA repair, I used a genome-editing technique called base editing to artificially introduce each point mutation into the genome of mammalian cells. After the desired modifications were verified via genome sequencing, the DNA repair outcomes inside individual cells were assessed using a fluorescence-based assay, flow cytometry, and my R script. Following these experiments, we realized that a previously unclassified point mutation should be designated as pathogenic because its DNA repair efficiency was reduced to compared to wild-type. Motivated to discover its biochemical mechanism, we employed AMBER molecular dynamics simulations and co-immunoprecipitation to probe changes in the mutant DNA repair protein's structure that may affect its interactions with its damaged DNA substrate and protein partners. Moreover, we used western blot and reverse transcriptase quantitative polymerase chain reaction to investigate changes in relative gene expression in the edited cells. Show less
