On a daily basis I prepare DNA libraries for Next Generation Sequencing in a High Throughput environment using Automated Liquid handlers like Agilent Bravo, Tecan, Echo and other instruments. Use Fragment Analyzer, Tapestation for QC of Dna libraries before loading them into Miseq I also prepare Highthroughput RT-qPCR and analyze gene expression data using current pipeline. My thesis project involves identifying oral microbes in the human plaque, salivary samples from healthy subjects, growing them on acrylic denture for comparing the bacteria in the patients with denture stomatitis, using genomic DNA extraction, gel electrophoresis, PCR amplification, 16srRNA sequencing methods, growing in the culture, media specific for candida albicans, streptococcus mutans, administering lactoferrin as a treatment, observing usnder scanning electron microscope.
