      Working in Respiratory Medicine at City hospital, with a focus on translational researchThis project utilises human lung cell samples to investigate profibrotic pathways in pulmonary fibrosis. In particular my work has focused on the avb6 pathway in epithelial cells and have intergrated clinical research into mechanistic studies. I was involved in the PROFILE study, a large collaborative study with GSK to look for genetic and blood bio-markers for disease. In addition, I have then undertaken preclinical work for novel anti-avb6 therapies, with Biogen.       My Ph.D project is focusing on developing a large animal model for pulmonary fibrosis to serve as a bridging model between what is found in murine studies and the human condition by providing a lung with similar structural and functional features to the human lung. Laboratory skills:- Handling of large animal for sampling i.e. blood, BAL, lung function - Collection and analysis of blood samples from large animals- performing lung bronchcoscopy for obtain BAL fluid,lung biopsies- Measuring and analysis lung functional data, currently using a bronchoscope - Dissection of lung and partitioning into appropriate samples for future analysis: formalin fixation, snap frozen, preparation for electron microscopy, isolation of parenchymal fibroblasts.- Processing and analysis of BAL fluid, differential blood cell counts- Cyrostat sectioning and preparation of frozen lung tissue for histology and immunohistochemistry- Histological and immunohistological staining of paraffin and frozen lung sections- Pathology interpretation and quantitative analysis of histology using Image J and Image Pro- Cell culture:o Aseptic handling of primary lung tissue for parenchymal fibroblasts- Protein analysis: Coomasie staining, SDS-PAGE, western blot, indirect ELISA, Metalloprotease activity via zymography- PCR: o mRNA extraction, Reverse Transcription, Real-Time PCR, RT-PCR array systemso interpreting and understanding mircoarray and RNA sequencing datasetsOther skills- GraphPad Prism 6, StepOne Software, Microsoft Word, Excel and Powerpoint- Time management- Student supervision- Can work to strict deadlines.- Self-motivated and highly curious - Great attention to detail- Problem solving skills, critical thinking and analytical skills.- Established and maintained effective collaborations- Oral communication to multidiscipline academics, and lay audiences- Written communication in the form of detailed and highly specific reports to lay summaries.
