      In this role, I was responsible for continuing some of the work started in my PhD project. Furthermore, I optimised a DNA sequencing assay to allow the detection of interleukin-33 DNA-binding sites. I routinely performed extracellular flux, stable isotope tracing (mass spectroscopy), PCR, immunoblotting, immunoprecipitation, proteomics, and electron microscopy, in addition to image processing/analysis, statistics and bioinformatics.       During this project, I identified the metabolic changes in retinal pigment epithelium cells during contexts of innate immune stress. I identified the protein AMP-activated kinase as a key energetic sensor which differentially regulates the metabolic adaptations to varying innate immune signals. In addition, have identified one immune signaling molecule (interleukin-33) as a metabolic checkpoint in retinal pigment epithelium cells, acting both through extracellular signaling and through internal transcriptional regulation of glycolytic enzymes and mitochondrial pyruvate oxidation.       This project involved the study of human eosinophil metabolism during quiescence and activation, understanding the differential metabolic response of eosinophils to immune signaling molecules between asthmatics and non-asthmatics. Techniques involved include primary cell culture, granulocyte isolation, extracellular flux analysis, immunosorbent assays, flow cytometry, cell viability assays, PCR and western blotting.       During this project, I investigated the responses of gestation-associated tissues (placenta, choriodecidua and amnion) to various innate immune stressors, assessing both cytokine output and key signaling pathways.
