      The subject of my MSc thesis was transport of sRNA between host plant and pathogen during which I mainly worked on gene cloning and transient expression assays in Nicotiana benthamiana. First, I have prepared knockout constructs of genes which were thought to be involved in virulence of Verticillium dahliae. Then, these were used to create V. dahliae knockouts by transformation of the spores in order to check whether these cultures would still be able to infect plants. Also, I studied targeting of the predicted virulence genes by a plant miRNA through transient transformation of N. benthamiana leaves. For that purpose, I have prepared constructs expressing the target gene tagged with a GFP and constructs expressing the miRNA itself. Then, when there is targeting a reduced GFP signal would be observed. Show less

      During this internship I have worked with plant cell and tissue culture to study gene expression changes during somatic embryogenesis, using transgenic fluorescent reporter lines in which candidate genes were tagged with fluorescent proteins. Target genes were involved in different steps of somatic embryogenesis such as genes required for hormonal signalling, root and shoot growth or embryo development. By following the expression of these genes using microscopy during embryogenesis a combining different fluorescent reporter genes I learned more about the genes which are involved in embryogenesis and how they affect each other. I have become experienced with stable transformation of Arabidopsis thaliana. I enjoyed this internship and learned a lot about early plant development. Also, during that internship and during my master thesis I have worked with gene cloning and plant transformation (stable transformation in A. thaliana and transient transformation in tobacco plants).
