I identified human-relevant bacterial proteins that alter the ability of E. coli cells to resist DNA damage from radiation exposure and studied the molecular mechanisms that drive these protective phenotypes. Major skills included: - Strain and vector engineering - Flow cytometry - Fluorescence microscopy - Mutagenesis I identified human-relevant bacterial proteins that alter the ability of E. coli cells to resist DNA damage from radiation exposure and studied the molecular mechanisms that drive these protective phenotypes. I studied the genetic regulation of cell-fate decision making processes in Bacillus subtilis. My dissertation focused on a novel link between the expression of the essential metabolic enzyme Acetyl CoA Carboxylase and the timing of sporulation initiation. I also contributed evidence for a new sporulation model that relies on internal kinase concentration rather than an unknown external starvation signal.Major skills included:- Strain and vector engineering- Protein expression and purification- Confocal and fluorescence microscopy- Nucleic acid isolation- Gel electrophoresis Show less
