▪ Designed and developed a novel modified CRISPR/Cas9 genome editing technology that greatly improve cell viability and reduce on-target and off-target INDELs, US/International patent WO2016205703A1 & WO2016205680A1 filed. ▪ Purified wild type Cas9 and over a 20 modified version of Cas9 recombinant proteins for RNP delivery in genome editing experiments. ▪ Successfully performed CRISPR/Cas9 enhanced gene correction of sickle cell disease(SCD) mutation in human sickle patient… Show more ▪ Designed and developed a novel modified CRISPR/Cas9 genome editing technology that greatly improve cell viability and reduce on-target and off-target INDELs, US/International patent WO2016205703A1 & WO2016205680A1 filed. ▪ Purified wild type Cas9 and over a 20 modified version of Cas9 recombinant proteins for RNP delivery in genome editing experiments. ▪ Successfully performed CRISPR/Cas9 enhanced gene correction of sickle cell disease(SCD) mutation in human sickle patient derived iPSCs and human/mouse sickle hematopoietic stem cells with high efficacy. Anayzed the short-term and long-term outcome of the genome editing using digital droplet PCR(ddPCR), sanger sequencing and next-generation sequencing technologies. ▪ Cured a humanized SCD mouse model by transplanting corrected hematopoietic stem cells using modified CRISPR/Cas9 system. 2nd and 3rd serial transplantation vaditated (First-in-the -world, unpublished data). ▪ Successfully purified a list of recombinant proteins and human/mouse cytokines for stem cell expansion and function study including bFGF, IL-6, c-Myc, TLR-9, etc.
