Research assistant with Dr. Jaan-Olle Andressoo's lab. Personally focusing on the effects of GDNF in schizophrenia, with the aim of developing a novel schizophrenia mouse model. Working together primarily with PhD candidate Daniel R. Garton. Wet lab techniques I utilised included: • Mouse genotyping. • RNA extraction; cDNA synthesis; qPCR analysis. • Western blotting; Amido Black staining; Silver Stain • Immunohistochemistry of cryostat mouse brain slices using an Apotome microscope • Dynabead Antibody Coupling; Synaptosome Extraction; Co-Immunoprecipitation of DAT from the synaptosome. • DAB immunostain • Stereology Animal techniques I developed in this job included: • General handling of mice • Sacrificing of mice • Cyclic voltammetry in live mouse brains • Use of anaesthesia/analgesia in mice • Mouse perfusion • Performing i.p./s.c. injection and oral gavage in mice and pups (including handling of methamphetamine, istradefylline and selpercatinib) • Home cage activity analysis for mice • Dissections in mouse brains and pup brains • Stereotax Injections of AAV to the striatum • Rotarod testing • Tail suspension test Office/team skills: • Statistical analysis using GraphPad software • Teaching lab techniques to MSc students • Oral/written presentations • Writing and editing of scientific manuscripts Show less

Grade: Distinction MSc Neuroscience Thesis Titled: Investigating the effects of BAF312 on demyelination and gliosis in Twitcher mice. Analysis of the effects of BAF312 was performed on cerebellar slices obtained from BAF312-treated Twitcher mice, the naturally occurring murine model for neonatal demyelinating disorder, Krabbes disease. Research was carried out in the hopes that BAF312 may prove a potential novel therapeutic for the treatment of Krabbes disease. Primary techniques included immunohistochemistry, using antibodies against markers for myelin, microglia, astrocytes and neurofilament proteins, and histological staining techniques such as luxol fast blue and periodic acid Schiff. Grade: 1.1 The title of my project was "The role of potassium ion channel inhibitors as a means to disrupt Alphaviruses such as Chikungunya, using Semliki Forest Virus as a model virus for the disease". This project included the use of skills such as cell culturing, cell proliferation/cytotoxicity assays, immunocytochemistry and Western blotting. 3T3 and BHK cells were used together with Semliki Forest Virus as the model virus and primarily KCl as the method of treatment.
