My primary role was to characterize the DNA binding requirements of BrxR, a putative transcriptional regulator that is linked to a subset of BREX defense systems. Later on, my research was focused on the BrxR homolog in Vibrio cholera Ban9. We did not detect specific binding, but we were able to partially restore binding by making a point mutation in the BrxR amino acid sequence. We hope to determine which amino acid positions may be significant in DNA recognition and how the BREX defense mechanism is regulated. Publication: Yvette Luyten, Deanna Hausman, Juliana Young, Lindsey A. Doyle, Natalia C. Ubilla-Rodriquez, Abigail R. Lambert, Kerilyn Higashi, Corina Arroyo, Kevin Forsberg, Richard M. Morgan, Barry L. Stoddard, Brett K. Kaiser. “Identification and characterization of the WYL BrxR protein and its gene as separable regulatory elements of a BREX phage restriction system.” Accepted for publication at Nucleic Acids Research. Presentations: “Characterization of BrxR, a putative transcriptional regulator in the BREX bacterial defense system” - Presented at Murdock Undergraduate Research Symposium, Vancouver, WA | November 2021 - Presented to potential Seattle University donors, Seattle, WA | April 2022 Lab Techniques: - PCR and primer design - Gibson cloning - Mutagenesis - Electrophoretic mobility shift assay (EMSA) - Ni-NTA protein purification - ApE and PyMOL analysis - Multiple Sequence Alignment (MSA)
