Pseudomonas aeruginosa evasion of neutrophil antibacterial functions in early Cystic Fibrosis lung infection • Extensive experience and knowledge in microbiology and mammalian cell cultivation • Develop high throughput functional assays to test the efficacy of therapeutic monoclonal antibody treatment against bacterial infection, eg. neutrophil bacterial infection, bacterial neutrophil adhesion assay, opsonophagocytosis killing assay (Collaboration with AstraZeneca) • Immunology: Perform various host cell and bacterial surface/ intracellular differential staining, acquire, and analyze samples by flow cytometer and flowJo; immunofluorescence: stain samples, acquire images using LSR confocal microscope and analyze them using image J or automated cell counting software Icy • In vivo animal models: familiar with various mouse models, including infection, handling, and processing of mouse organs, as well as other pig and wound infection models • Biochemistry: quantify bacterial or host cell proteins using immunoblotting, ELISA • Molecular biology: nucleotide extraction (DNA, RNA and Plasmid), cDNA synthesis, RT-qPCR, molecular cloning, Gibson cloning technology, primer design, PCR, gel electrophoresis, restriction enzyme digest, DNA ligation, promoter-reporter design and expression measurement • Cell purification and isolation: isolate and purify both primary human and mouse neutrophils • Virology: isolated and worked with bacterial phages, plaque assay • Supervisor: mentored and trained undergraduate and graduate students • Teamwork: assisted with colleagues’ research projects and collaborated with multiple international external partners • Scientific writing: published in peer-reviewed journals, thesis, conducted literature reviews, and reviewed research articles with supervisor • Scientific evaluation: data analysis and experimental troubleshooting Show less
