ABSTRACT OF MSc Regulation and control of the cell cycle is crucial for growth and development of eukaryotic organisms. Specific degradation of proteins via ubiquitin/26S proteasome system (UPS) is one of the requirements for the progression through the cell cycle phases. Ubiquitin post-transductionally modifies target proteins, tagging them for the degradation in 26S proteasome. Sequential action of three enzymes, E1-activating enzyme, E2-conjugating enzyme and E3 ubiquitin ligase is necessary for the Ub ligation. There are different types of E3. One of them is SCF complex, composed of four subunits: Cullin, RBX, SKP2 (ASK1 in plants) and F-box protein. Target specificity of the SCF complexes depends on the F-box proteins. Based on homology to the human SKP2, two F-box proteins, SKP2A and SKP2B, were identified in Arabidopsis, a model plant. These two F-box proteins seem to be involved in regulating cell cycle in Arabidopsis. SKP2A interacts with members of ASK family and CUL1, forming SCFSKP2A complex with E3 ubiquitin positively regulates cell cycle by degrading E2FC/DPB transcription repressor. In addition, SKP2A is also degraded by the Ub/26S proteasome pathway in an auxin depentent manner. In a recent report, it has been shown that SKP2A binds specifically and directly to auxin, suggesting that this F-box protein might be a novel auxin receptor.
