The Role:- carrying out selection of aptamers against small molecules independently (both automated and manual selection);- carrying out pre-tests for new targets (solubility and stability study, PCR test);- production and quality control of single stranded DNA pool for further selection rounds using following techniques: asymmetric PCR, lambda exonuclease and exonuclease I, automated and manual bead purification;- recording results, analysing and initial interpreting for further review by Line Manager,- providing scientific and technical support for developing original solutions to problems, improving existing methodologies/protocols and troubleshooting;- supervising and training junior members of staff where appropriate;- effectively scheduling laboratory work to ensure timely completion of required tasks;- facilitating capture bead production, bead regenertion and quality control;- preparing primers’ stocks and buffers for small molecules department;- ensuring all paperwork is completed accurately and in a timely manner – protocols, reports, and summaries in both electronic and paper formats;- working in an orderly way – keeping the laboratory work area safe and clean;- supporting Good Laboratory Practice implementation and maintenance. Stem Cells Group During the internship I had the opportunity to understand the methods used to inestigate the molecular mechanism of retinal diseases. The project that I've been helping with is based on using CRISPR/Cas9-iPSC (induced Pluripotent Stem Cells) as an excelent model to study the most common cause of blidness - Retinitis pigmentosa. Experiments that I carried out involed techniques like PCR, Real Time PCR and surveyor assay to characterize CRISPR/Cas9 colonies. Institute of Environmental Mutagenesis Institute of Human Genetics Polish Academy of Sciences Supervisor: dr Maciej Giefing The goal of the project, which I have been a part of, was identification of mutations in SDE2 i DMRT1 genes, which may be connected with tumouring. During the internship I have became familiar with useful laboratory methods such as Fluorescence In-situ Hybridization (FISH) and bacterial artificial chromosomes (BAC). I’ve also learned how to work with human cell lines in antiseptic conditions. Show less
