Analysis of intracellular pH changes of yeast (Saccharomyces cerevisiae) cells upon changes in extracellular pH and external potassium concentration. Measurements of intracellular pH using cells expressing the gene encoding for pHluorin, a GFP (green fluorescent protein) variant that changes its fluorescence properties depending on pH. Implementation of the FLISE (flux determination using ion-selective electrodes) method to determine potassium ion flux.
