      Topic: Epigenetic modifications in the anticodon region of tRNAs fine-tune its stability and functionality. The modification N6-threonylcarbamoyladenosine (t6A) at position 37, adjacent to the anticodon stem-loop in tRNA, is well understood, however the cyclization to ct6A remains elusive. Threonylcarbamoyladenosine dehydratases (Tcd) have been proposed as crucial players for the enzyme-dependent dehydration of t6A to ct6A, but their structural and biochemical characterization is missing. Methodological focus: Construct design, molecular biology methods (PCR, cloning, mutagenesis, transformation in bacteria and yeast, plasmid preparation), production of knockouts in yeast, in vivo tagging of yeast genes, protein expression (E. coli and yeast) and purification (IMAC, SEC, IEX), ESI full-length MS for protein verification, structural protein characterization (HPLC, AUC, SAXS), SDS-PAGE (Coomassie staining), EMSA for RNA detection (SYBRGold staining), in vitro transcription of tRNAs, tRNA sequencing, Thermofluor assay, Western blotting Main focus: Crystallography ( handling of crystallization robots, selection of crystallisation conditions, seeding, crystal fishing, measurement of data at PSI, data evaluation) Collaboration with other chairs in the fields of electron microscopy, SAXS, NMR, AUC, NGS, mass spectroscopy Used Software: Snapgene, Unicorn(Äkta), Gimp, Pymol, CorelDraw, Coot, CCP4, Supervision of trainees, study-relevant research internships as well as practical courses Peer review of manuscripts in scientific journals Show less
