Center for Musculoskeletal Research - Dr. Chao Xie Lab Project 1: Targeted Antibiotics to Treat Osteomyelitis • Designed and tested the impact of bisphosphonate conjugated antibiotics on S. aureus mediated bone infections in murine models, significantly reducing bacterial bone loss by 50% • Reconstructed 100+ micro-CT images with Amira to create a 3D model of infected bones and quantify loss • Performed aseptic surgery to implant infected pins in murine femurs and conducted… Show more Project 1: Targeted Antibiotics to Treat Osteomyelitis • Designed and tested the impact of bisphosphonate conjugated antibiotics on S. aureus mediated bone infections in murine models, significantly reducing bacterial bone loss by 50% • Reconstructed 100+ micro-CT images with Amira to create a 3D model of infected bones and quantify loss • Performed aseptic surgery to implant infected pins in murine femurs and conducted histological analysis • Conducted bacterial colony formation unit counting assays, collected and homogenized infected tissue Project 2: Exploration of Inflammation Cell Markers • Studied the role of CD-163 in fracture healing and angiogenesis and found that inhibiting CD-163 significantly reduced blood vessel formation by 30% making it a poor drug target despite its anti-inflammatory abilities • Captured x-rays and bioluminescent images and quantified cell densities from histological images using ImageJ • Analyzed the stress-strain biomechanics of bone in CD 163-/- mice, finding a ~20% decrease in yield stress • Maintained CD 163-/- mouse colonies and performed PCR genotyping on mice Project 3: Bacterial Biomaterial Interaction • Examined the ‘race for the surface’ between host immune cells and S. aureus on biomaterials • Utilized Imaris and ImageJ to manipulate multiphoton images to generate 3D visualizations of macrophages, neutrophils and S. aureus and produce volumetric quantifications • Assisted in the surgical implantation of a grin lens aperture in infected murine models for multiphoton imaging • Cultured human macrophages and labeled the cells fluorescently for phagocytotic analysis Project 4: Disrupting Infections with Monoclonal Antibodies • Examined the effect of TPH 101 antibodies on S. aureus infection pathology in murine models • Collect blood from mice and separated supernatant from blood samples for antibody analysis
