  Used a wide variety of experimental procedures including western blotting, transfection, transformation, and immunostaining to analyze how mouse embryonic fibroblasts null for the gene KLF4 react when the gene is reintroduced and expressed in cells.Specifically I transfected a plasmid containing GFP and KLF4 controlled by a tetracycline on/off system into KLF4 null MEF's and examined how reintroduction affected protein levels of cyclin D, cyclin E, p21 and p53 using Western Blotting.I used immunostaining to examine how reintroduction of KLF4 regulated the centrosome cycle and whether KLF4 introduction reduced the levels of cyclin E and reduced centrosome amplification, a marker that indicated genomic stability.
