Syracuse, New York Area Developed and optimized the Friedel Craft’s reaction of indoles with trichloroacetimidates. Discovered the effects of Steric bulk in trichloroacetimidate moiety on regioselectivity of these reactions. Studied alkylation of 3-substituted indole derivatives and tryptamines with trichloroacetimidates which provided practical opportunity for rapid construction of pyrrolidino[2,3-b]indoline. Investigated N-Alkylation of 2,3 disubstituted indoles and their rearrangement to furnish C-3 alkylated compounds. Introduced new technology for protection of carboxylic acids under relatively neutral reaction conditions using 4-methoxybenzyl trichloroacetimidate and diphenylmethyl trichloroacetimidate. Designed, modified and optimized multiple synthetic routes for efficient and safe scale up of lead molecule, which met initial requirements for Tryptamine based SHIP inhibitor. Developed a diverse library of SHIP inhibitors.
