Currently, I am working towards developing single molecule fluorescence based DNA sequencing technique which will have the ability to read and identify sequence as short as five nucleotides. We shall apply this technique to understand various aspects of transcription process. I have worked on elucidating the molecular mechanism of hearing using biophysical approach. For this work, I extensively use recombinant protein expression using both bacterial and mammalian cell lines, recombinant DNA technology, CRISPR-CAS technique to alter genes, protein purification using chromatography. Plethora of biophysical instruments have been used to characterize protein like analytical ultracentrifugation, DLS, steady state and time-resolved fluorescence, ITC, DSC, single molecule FRET using TIRF, AFM based force measurements. I have also performed extensive molecular dynamics simulation to look into the atomic level changes in protein. Show less
