Given that… Show more On December 2022, I presented my proposal for an independent research project in the Iwase lab in hopes to write an honors thesis for my neuroscience major and began my project in January 2023. The Iwase Lab focuses on how histone-modifying enzymes regulate brain development and how their dysfunction results in neurodevelopmental disorders, focusing specifically on KDM5C, a histone 3 lysine 4 demethylase whose dysfunction results in X-linked mental retardation disorders. Given that demethylating trimethylated H3K4 requires alpha-ketoglutarate as a co-substrate, my project explores the potential regulatory dynamics of nuclear metabolic flux on the activity of alpha-KG-dependent epigenetic modifiers, specifically over the course of LTP induction and the role that may play in long-term memory formation. At this stage, my work over the last few months has been dedicated primarily to the development of an in vitro model in primary mice neuronal cultures that can report changes in nuclear alpha-KG concentrations by fluorescent, ectopically expressed reporters. I have also done minor experiments with immunofluorescence to track the nuclear localization of metabolic enzymes in neurons that can locally supply alpha-KG as a reaction product to epigenetic modifiers upon neuron stimulation.
