•ONCR-021 (CVA21-LNP): -Designed and optimized new ribozymes and leaders to achieve maximum linealized ONCR-021 RNA during manufacturing. -Developed in-vitro assays to quickly assess kickoff and screen different ONCR-021 ribozymes and leaders. -Validated receptor necessity and sufficiency of ONCR-021 in human cancer cell lines. -Determined cross-species specificity of ONCR-021 viral entry receptor. -Characterized ONCR-021 viral genomic drift rate (replication fidelity). -Characterized ONCR-021 RdRp recombination rate. •ONCR-GBM (EGFR-targeted HSV1): -Demonstrated ONCR-GBM entry receptor targeting from Nectin-1 only to EGFR and Nectin-1 -Validated ONCR-GBM EGFR receptor necessity and sufficiency in human GBM cell lines. •New Discovery candidates (vRNA-LNP): -Developed and validated 3 novel oncolytic synthetic viruses as candidates for our IV self-amplifying vRNA-LNP platform. -Optimized IVT RNA template to completely remove abortive IVT RNA production. -Optimized IVT RNA capping for maximal kickoff and suitability for large scale LNP-RNA encapsidation. -Optimized the design, production and purification of numerous viral vectors and lentivirus vectors (LVV). -Armed the new viruses with therapeutic immunostimulatory payloads and optimized payload production to higher levels than for our other platforms. -Experience with creation and optimization of chimeric RNA viruses with different entry receptor tropisms. -Implemented a stable IRES-dependent strategy to limit possible community transmission of our new candidates. -Performed in-vitro selection of best indications and syngeneic lines for future candidate development. -Mentored and trained numerous co-ops, students and lab personnel.
