Use of yeast in combination with available pharmacological tools to investigate the G Protein-Coupled Receptor, GPR55. The aim is to determine the functional effect of different antagonists in yeas-based assays and to estimate their affinity and selectivity to GPR55 as well as hypothesising their mode of action. Assessment of their activity at roedent GPR55 was also carried out. Bacteria followed by yeast transformations were performed to get the yeast strains ready for the experiments. Finally, flow cytometry technique as well as the most potent GPR55 antagonists previously studied were used to reveal GPR55 function in neutrophils. Show less
