By combining experimental and computational approaches, we assessed the conformational ensemble changes in p53 TAD resulting from… Show more Project 1: STRUCTURAL AND FUNCTIONAL CONSEQUENCES OF THE CANCER-LINKED MUTATIONS IN THE TRANSACTIVATION DOMAIN (TAD) OF P53 My research focused on understanding how the cancer-linked mutations in the N-terminal transactivation domain of p53 could perturb the balance between the binding of p53 to Human-Double-minute-2 (HDM2) and CREB-binding protein (CBP). By combining experimental and computational approaches, we assessed the conformational ensemble changes in p53 TAD resulting from the cancer-linked mutations. (NMR and Molecular simulation approaches were incorporated by collaborations) Project 2: SKD3/HUMAN CLPB, AN AAA+ ATPASE LINKED TO MULTIPLE PATHOLOGIES: CELLULAR LOCALIZATION AND PROCESSING SKD3, also known as mammalian ClpB, belongs to the AAA+ family of ATPases associated with different activities. The actual function of SKD3 is unknown. Mutations in the skd3/hclpB gene have been linked to various pathologies with a broad phenotypic spectrum, including 3-methylglutaconic aciduria, cataract, intellectual disability/developmental delay, congenital neutropenia, progressive brain atrophy, and movement disorder. The clinical manifestations of the mutations are currently impossible to explain due to the lack of understanding of the biological function of SKD3. Our goal of this project was to search for interacting partners and substrates of SKD3/hClpB and investigate how the ATP-dependent function of SKD3/hClpB supports the mitochondrial biogenesis, metabolism and/or proteostasis.
