Curricular internship in Signal Transduction (Jerónimo Bravo's research group). Worked together with a recent doctorate in the group until I was tasked with the cloning and heterologous protein expression of a ribosomal assembly factor. Applied several molecular biology and biochemistry techniques such as: ● PCR and denaturing polyacrylamide gel protein electrophoresis. ● DNA extraction and agarose gel electrophoresis. ● Ligation-independent cloning. ● Chemically competent E. coli transformation and culture. ● Protein expression using autoinduction and IPTG induction. ● Protein purification via immobilized metal affinity chromatography with nickel resin and glutathione affinity chromatography. ● Directed mutagenesis. Extracurricular internship in Metabolic Growth Signals and Regenerative Medicine (Luke Noon's research group). Participated in the different lines of research, including helping with the screening of CRISPR/Cas9 clones for gene knockout. Was made responsible for carrying out a series of experiments, first reproducing former findings, and then researching and designing new protocols, and presenting the data in a lab meeting. Developed skills in experimental design, scientific communication and data analysis and became accustomed to the techniques used, which included: ● Cell culture. ● RNA extraction. ● qRT-PCR. ● Time-lapse microscopy. ● Agarose gel electrophoresis.
