As a research assistant I colaborate in the projects developed in Dra. Laura Sanz Alcóbers's research group. Within the different projects I colaborate with, should be highlighted the generation of CRC organoids for immunotherapy research and the analysis of the effects of Wnt inhibitors in the behaviour of CRC tumor lines. Experience in cell culture, organoid culture, flow cytometry, cytotoxicity assays, cellular transduction, lentiviral handling. As a research assistant I colaborate in the projects developed in Dra. During my Major Internship in Emile Voest group I have analyzed how inter- and intra-tumor heterogeneity can lead to differential sensitivity against autologous T cells, that will result in a specialized treatment. The main techniques that I performed during this internships were: Organoids culture, T cell culture, Organoids – T cell co-culture, Killing Assays, Flow Cytometry, FACS, Organoids transduction, CRISPR-Cas9, FlowJo. Under the supervision of Yvette van Kooyk's group I have written a review on the therapeutic purposes of in vivo monocyte-derived dendritic cells vaccination for cancer treatment. For this review I adressed the main questions about how monocytes can be attracted to the vaccination site, how their differentiation into MoDCs can be induced and how MoDCs can promote anti-tumoral immune response. During my Minor Internship in the Netherlands Cancer Institute (NKI) I performed Immunofluorescence combined with Fluorescence In Situ Hybridization (IF-FISH) in order to study de effects on telomeres' maintenance upon ggNER and TLS depletion. Cell culture, confocal microscopy, imaging analysis and quantification with FIJI and IMARIS are other techniques that I also performed during my internship in the NKI. During my Bachelor Degree in Biotechnology I did my internships in the Hospital 12 de Octubre performing Sanger sequencing and Multiplex Ligation-dependent Probe Amplification (MLPA) in gliomas' samples in order to identify mutations in the ATRX gene and demonstrate that the immunohistochemical analysis always has to be accomplished with a genetic-molecular analysis to prevent false positives or false negatives.
