Under the Supervision of my Research Mentor, I worked on the Cloning of Acetate Kinase in Escherichia coli. I learned many skills and techniques such as PCR, restriction enzyme digestion, ligation, Gel Electrophoresis, preparation of complex buffers, design of primers, DNA Isolation, aseptic techniques, transformation via heat shock as well as the use of machines such as Nanodrops and other spectrophotometers. We were able to successfully clone and transform E. coli with pColdII vector containing the Acetate Kinase Insert.
