Thesis: Generating anti- human CD45 monoclonal antibody to using in blood disease diagnosisAbstract: Monoclonal antibodies have been applied in many laboratory diagnoses of various diseases, especially in cancer diseases because of their specificity in interactions with certain cells or proteins. Anti- human CD45 monoclonal antibody has been popularly used in diagnostics of leukocyte diseases, such as leukemia and myelosuppresion,… using flow cytometry method as CD45 is a leukocyte common antigen. In this project, we generated anti-human CD45 monoclonal antibody to satisfy the domestic diagnostic demand. Recombinant huCD45 antigen was expressed in E. coli BL21 (DE3) and purified using affinity chromatography. The purified recombinant huCD45 antigen was then injected to Balb/c. The spleen cells from immunized mice having the highest specific antibody titer were collected and fused with myeloma to generate hybridoma. Hybridoma cell line secreting highest yield of anti huCD45 antibody was cultured. The secreted anti- CD45 antibody was purified using protein G. The properties of the anti CD45 antibody were tested. This antibody is IgG2b/ isotype; has the dissociation constant: Kd = 7,13 ±1,01×10-10 (M); it can interact with recombinant huCD45 antigen through ELISA test and western blot; Especially, this anti huCD45 monoclonal antibody can recognize the natural CD45 antigen expressing on the surface of JurkatCD45+ cell in Western blot and fluorescent immunostaining test. Therefore, the anti CD45 antibody generated in this project has high affinity with CD45 antigen and it is available in a variety of antibody-based applications including ELISA, Western Blot, and immunostaining.
