My project in the Morrissey Lab involved manipulating membrane receptor clusters to study phagocytic signaling, specifically, how clustering of CD47 and SIRPa affect phagocytosis. CD47 is an inhibitory signal ubiquitously expressed on plasma membranes of healthy cells. CD47 binds SIRPa on macrophages, initiating an inhibitory signaling pathway that prevents phagocytosis of CD47-expressing cells. To directly manipulate clustering of CD47 and SIRPa, we cloned multiple CD47-CRY2 constructs that cluster upon light stimulation. Show less
