Gajraj can design and carry out his experiments in a scientifically correct manner. He gives utmost care while carrying the analysis and the preparation of samples.       I was a research intern in the Drug Product Development team wherein I routinely worked with analytical techniques such as Protein A-HPLC, Cation Exchange Chromatography,Capillary Electrophoresis, SEC-HPLC, Endosafe PTS for purification and characterization of antibody samples. I also worked on method development for analysing sialic acid in glycoproteins using UPLC-FLR (Ultra Performance Liquid Chromatography-Fluorescence Detector).       During my master thesis at the University of Geneva, I worked on the non-invasive topical delivery of Infliximab for the treatment of psoriasis. We developed ELISA for antibody quantification which was validated for its accuracy and precision. It gave me insight into ICH validation guidelines for analytical method development. For antibody-drug delivery, our focus was on using iontophoresis for reaching deposition level above the therapeutically relevant value of 100xEC50. We employed the cutaneous bio-distribution method for precise drug profiling in the skin and were able to develop an iontophoretic patch prototype. The process of device development was very intriguing and gave me the opportunity to work in an inter-disciplinary environment. The prototype will be used for carrying out topical delivery in vivo, planned for later 2021. Show less

      As a Research Assistant at the Institute of Molecular and Cell Biology, my role was to perform in-vivo studies in mice (BALB/c) with the aim of elucidating the molecular mechanisms involved in immune checkpoint inhibition, a popular approach in cancer immunotherapy. I was directly involved in experimental design which involved injection of tumor cell lines (Syngeneic cell lines) in mice and monitoring their growth over time. Immune profiling was performed on the tumor extracted after 14 days, through CyTOF mass cytometer. We further employed the use of PD-1 inhibitor antibody to view its effects on tumor growth. The results were promising as the tumor volume showed significant reduction after drug treatment. During the project, I gained hands on experience with state-of-the-art techniques such as LC-MS/MS, which was used for pharmacokinetic studies and it was my first experience of working in pre-clinical environment which further motivated me to work towards translational research.       The aim of the project was to determine the role of caspases in inducing apoptosis through the activation of TAF6δ, an inducible pro-apoptotic subunit of the general RNA polymerase II transcription factor, TFIID. I tested multiple cell lines(HeLa, Jurkat, MDA-MB-231, HL-60) for their response to inducers of apoptosis upon caspase cleavage of model substrates. During the course of this project, I mastered mammalian cell culture, protein extraction and quantification and Western Blotting, all of which were extensively used throughout my internship.
