Research project: Synthesis and characterization of small chiral organic molecules to be used as metal-free catalysts in different organic stereoselective transformations; their support on different materials (silica nanoparticles, soluble polymers i.e. PEG, non-soluble polymers i.e. polystyrene, silicon wafer or glass slide) through grafting or other wet chemical techniques. Title of thesis: “Pd and Pt nanoparticles synthesised by lignin: characterisation, catalytic application and biocompatibility.” ・ The research activity, framed in the context of “green chemistry”, deals with the synthesis of new homogeneous and stable metal nanoparticles (Pt and Pd NPs) made in water by lignins and fulvic acid. The natural polymers act as reducing and stabilizing agents. For NPs characterization, UV-Vis spectroscopy, XRD, NMR, TEM, IR were used. Such NPs were also tested as catalysts in alcohols oxidation, in some reductions (p-nitrophenol, allyl alcohols, unsaturated ketones or azide) and in C-C cross coupling reactions. ・ March 2012-October 2012: period spent abroad for a collaboration with “Biocatalysis and Organic chemistry” group, University of Technology Delft (NL). Tutor: Prof. I.W.C.E. Arends and Prof. F. Hollman Department of “Biocatalysis and Organic chemistry”, University of Technology Delft (NL). The research was focused on the realization of chemo-enzymatic catalyzed one pot cascade reactions. Studies of mutual inhibition between NPs and alcohol dehydrogenase or cromatoreductase were carried out. It was observed that NPs shape and size are the critical parameters for the toxic effect on enzyme. Some methods to avoid direct binding of NPs with the protein were tested (sol gel, silica encapsulation, NPs in ferritin). Measurements of enzymatic activity in the presence of NPs and attempted characterization of NPs–bioconjugates formed in situ were made using UV-Vis spectroscopy and DLS, XRD, size-exclusion chromatography-HPLC, circular dichroism.
