      IgA deficiency (IgAD) is the most common primary immunodeficiency among Caucasians. Still, the lack of IgA does not always result in clinical symptoms and only a subset of sIgAD patients suffers from recurrent infections, autoimmune diseases or develop other immune deficiencies. Here, we questioned whether IgA-expressing B cells can be found in sIgAD patients and whether their IGHV regions carry normal signs of antibody maturation. Symptomatic IgAD patients have impaired IgA B-cell memory from both local intestinal (CD27-) and germinal center-derived responses. Still, this is restricted to a quantitative effect, because SHM seemed normal in sIgAD patients. The decreased IgA2 usage in patients could indicate switch defects to downstream constant regions. However, this is specific for IgA, because IgG transcripts showed increased switching to the downstream IgG2 region. Show less

      Cloning of resistant gene analogue from pearl millet against downy mildew In recent years, there has been evidence of severe loss and destruction of Pearl Millet crop and decrease in yield significantly. The causative organism, Sclerosporagraminicola is a soilborne fungus which causes downy mildew disease in the plant. Most plants tend to show a type of innate immunity towards specific diseases by the presence of resistance (R) genes in their genome, which is expressed at the time of pathogen attack. These R genes are specifically characterized by a NBS-LRR domain. Based on sequence information of the conserved domains can be invariably used to mine similar sequences from other plant species using degenerate and specific primers for their amplification in PCR. We the species specific primers were designed for amplification of desired amplicon and this was further cloned in E.Coli JM109 host using cloning vector pTZ57R/T, a 2886bp circular plasmid. Upon blue-white screening, the desired amplicon was isolated from the white colonies and further cloned into E.Coli JM109 host using expression vector pRSETA, a 3Kb linear plasmid. Analysis of transformed cells after colony PCR by agarose gel electrophoresis confirmed the introduction of desired amplicon into expression vector. Show less
