I have demonstrated that a subfamily of tetraspanins (molecular organizers of the plasma membrane), that we called TspanC8, can directly interact with the metalloprotease ADAM10 and regulate its trafficking, activity and specificity towards its Notch substrate. I finally demonstrated that TspanC8 constitutes a new family of Notch regulators with evolutionary conserved functions in the regulation of ADAM10 trafficking and activity, as Drosophila TspanC8s were shown to regulate Kuzbanian (ADAM10 ortholog in Drosophila) activity.
