Our work demonstrated bidirectional promoter activity inherent to eukaryotic promoters within genome inserted cassettes caused this off-target effect, thus providing a means to prevent it in future studies. We then designed and shared an improved set of selection cassettes for use in Saccharomyces cerevisiae which prevent this off target effect.
