Research Engineer, Manager of the Biotechnology part of the REALCAT platform in Lille. REALCAT is a technical platform integrating some 30 high-throughput equipment in the field of Biotechnology and Chemistry. Our core business is Catalysis, whether Biocatalysis (enzymatic and fermentative), Chemical Catalysis (homogeneous and heterogeneous), or Hybrid Catalysis. Although very diversified, our projects mainly concern the valorization of Biomass in a wide range of applications, ranging from new generation biofuels, to the synthesis of innovative polymers, as well as the production of platform molecules for industry or the search for new antimicrobials. Personally, my job is to manage, coordinate, and carry out all the projects in the Biotechnology part of the platform. My PhD thesis is titled: "Seeking New Transaminases for Chiral Amine Synthesis." PhD realized under the supervision of Prof. Thierry Gefflaut and Dr. Franck Charmantray. Description: My work consisted in the development and implementation of new high throughput screening biochemical assay for the discovery and study of new enzymes. The two main types of enzymes I've worked with were Transaminases and Monoamines Oxidases. To carry out my project, I first had to develop assays at the chemical and biocatalytic level (synthesis and preparation of substrates, selection of enzymes, design of model reactions, determination of optimal conditions, etc.). The second step consisted in the evaluation of all the statistical parameters concerned (blanks, negative controls, detection thresholds, LOD, selection thresholds, etc.). It was then necessary, during the third step, to proceed with the automation of the tests (programming of a pipetting platform / TECAN detection, writing of computer programs for the automatic analysis of the screening results) in order to be able to use them in high throughput screenings. Finally, I was able, in a last step, to carry out the screening of a 2000 Transaminases library (built in partnership with the Génoscope (Evry)) to find and study new biocatalysts potentially useful for therapeutic target synthesis.
