  The Ph.D. project assigned to me aims to prepare human membrane proteins for structural and interaction analysis. Molecular cloning and transformation/transfection of Escherichia coli cells, Sf9 insect cells and HEK293T cells of recombinant vectors are being conducted in order to find the proper solution of membrane protein over-expression. Additionally, recombinant baculoviruses are being produced to infect Sf9 cells in small and large-scale. Moreover, high-performance liquid chromatography (affinity, ion exchange and size-exclusion chromatographies) is being applied to prepare high-quality protein preparation, tested by SDS-PAGE and Western Blot. In the end, samples are analyzed by Cryogenic Electron Microscopy. The experimental plan is designed and applied by the Ph.D. candidate himself with help of a Post-Doc researcher and the Supervisor (Prof. Dr. Irmi Sinning). Show less

  Characterization of the dynamics of Biomolecular Condensates in Myofibrillogenesis. The project is conducted entirely in vitro via cloning, by over-expressing and by purifying recombinant proteins in E. coli, applying affinity, ion exchange and size-exclusion chromatographies (AKTA system). Later, those proteins are labelled with fluorophores using Sortase-A reaction or they are fused to natural fluorophores, like GFP. Further, proteins are imaged with Spinning Disk Microscope and Differential Interference Contrast Microscope and their dynamics is studied applying Fluorescence Recovery After Photobleaching. Data processing is conducted using ImageJ and GraphPad (experienced user), producing images and time-lapse and 3D videos.
