      • Working in the Balchin lab on protein biogenesis • Significantly enhanced my experimental and analytical skills through completing HDX-mass spectrometry, circular dichroism, DNA and protein purification, site-directed mutagenesis, gel electrophoresis, western blotting and enzyme assays • Improved my presentation skills through progress reports and presenting scientific journals Trigger Factor (TF) is an ATP-independent bacterial chaperone which promotes protein folding. It is the only bacterial chaperone known to interact with the ribosome exit tunnel during protein translation. The most extensively studied TF is from Escherichia coli (E. coli) which has structured domains, but lacks long disordered extensions. Interestingly, sequence analysis and AlphaFold predictions of the structure of TFs from different species have revealed disordered extensions at the C-terminus close to the substrate-binding domain, but these extensions are not conserved throughout all bacterial species. Nevertheless, eukaryotic TFs found in chloroplasts have disordered extensions, as seen in TF from Arabidopsis thaliana. My research project aims to determine whether the disordered extensions in TF contribute to its chaperone activity. Show less
