      Project: "Structural determinants of activity and non-canonical regulatory mechanisms of Protein Kinase A (PKA) in sperm physiology" Location: Cellular-Structural Biology Lab (coordinated by Andrés Binolfi, PhD) and Cell signal transduction networks (coordinated by Darío Krapf, PhD) Summary: PKA is a kinase enzyme that acts as a central coordinator of the sperm capacitation process. Previous work suggests that the regulation of this protein is complex and multifactorial, involving post translational modifications, such as acetylation, as well as active restriction of its subcellular localization. By using both in cellulo and in vitro studies with a recombinant version of PKA, we are investigating non-canonical regulation mechanisms for this enzyme. Show less

In particular, I studied the role of PKA and Src enzymes, and their association with different capacitation events. In a collaborative work, we developed a method to predict the outcome of Assisted Reproductive Techonologies (ART) by assessing sperm samples from male patients. Show less

      Project: “Design and characterization of a hybrid protein between matallo-β-lactamase of types B1 and B2” Location: Metalloproteins Lab Summary: metallo-β-lactamases are enzymes produced by different bacterial strains, that provide resistance to β-lactamic antibiotics (penicillins, cephalosporins, carbapenems). Such enzymes can be further sub-classified in three groups, each of them with different kinetic properties and substrate affinity. In order to identify structural elements that modulate substrate specificity, we designed, expressed, purified and characterized a chimeric enzyme that combines features of both metallo-β-lactamases B1 and metallo-β-lactamases B2.
