• Designed and generated 62 positional libraries of an RNA binding protein and interpreted its sequence-functional landscape via Next Generation Sequencing (NGS). Received presentation opportunities at multiple platforms, including in Gordon, ACS, and Protein Society • Optimized high-throughput expression and purification of the positional libraries and identified multiple point-mutants with thermal stability higher than WT• Identified amino acids responsible for the binding of notch ligand by protein sequence and structural analysis• Optimized large-scale expression and purification protocol of notch ligands in the bacterial system to achieve 5x higher yield and obtained 2x higher resolution NMR• Cloned and established mammalian expression system in Expi 293F cell line for notch ligands to determine binding affinity using SPR• Coached six PhD. students in experiment design, lab techniques and data interpretation• Mentored four undergraduates in acquiring biochemistry lab techniques, designing experiments, writing abstracts and completing poster presentations Show less
