Our research is directed at identifying novel post-translationally modified proteins in C. elegans and elucidating their biological function. As an alternative to radiolabeling, we utilize azide- and alkyne-sugar analogs which are compatible with downstream ‘click-chemistry’, affinity purification and proteomics. I have extended this strategy by developing a novel method to detect phosphorylcholine protein modifications, which are implicated in immunomodulation and host evasion by parasitic nematodes, by metabolically labeling proteins with an alkyne modified choline analog.
