Optimized the cell line and number, incubation time, plating conditions, and the stress used to induce phosphorylation of the target, resulting in the creation on an optimized protocol used weekly in funnel to screen compounds. • Advanced an anti-cd3e model in C57BL6 vs. Balb/C mice critical for screening funnels; enhanced both models regarding incubation times, cell numbers, reagent concentration and analysis, incorporating this into another weekly protocol. • Re-established a cellular assay that had stopped working due to weekly variances in the target enzyme in the decided cell line. After testing different cell lines with different levels of and types of stress concluded the original cell line was the best. The stress method of induction was abandoned, and a stably transduced version of the cell line that expresses increased levels of kinase was created.
