One part of my mission is to improve CRISPR-Cas-9 technology to establish new mutants of nematode using this technique. Until now I managed to obtain several mutants using CRISP/Cas-9 and Mos techniques. I also lead several genetic screens to identify novel genes involved in the formation and the regulation of synaptic functions in the nematode. I also isolated several interesting mutants after EMS mutagenesis which have been sent to whole genome sequencing. They are currently being characterized more in details. *Genome engineering and moleclular biology: - Site directed mutagenesis of nematodes using: CRISPR/Cas-9, Co-CRISPR, MosSCI and MosTIC techniques - Restriction cloning, GIBSON cloning, Gateway cloning. - Microinjection into the gonade of nematode. *Detection of GABAergics or cholinergics receptors mutants followed by characterization - Screening planification - EMS mutagenesis - Visual screen using macrozoom - Detection, characterization and registration of mutants - Fixation and Immunostaining of nematode - Spinning Disk microscopy - gDNA prepration for Whole Genome Sequencing. *Supervision of a Master 2 student Responsabilities: - Manage lab's stock - Update lab's database for plasmid, primers, strains - Relation with sales representatives Show less

I had to develop new transgenesis tools for several nematode species. I also studied mitotic spindle diversity among nematode species using DIC microscopy. This work led to the publication of one scientific article and an other one will be publised. *Development of transgenesis methods for other nematode species - Adapation of a common transgenesis method used in C. elegans to genetically modify other nematode species. - Begin to develop a new transgenesis method (TALEN). - Biolistic bombardment of nematode to insert several copies of a transgene. - Microinjection into the gonade of nematode *Molecular biology expertise - Development of RNA synthesis and purification to microinject nematodes. - RNA extraction and RT-PCR - Restriction cloning, Gateway cloning - Freezing of worms and update of lab’s databases (nematode strains, plasmid, primers) *DIC microscopy and statistical analysis - DIC microscopy recording of the first mitotic division in nematode embryos among more than hundred strains. - Use of an algorithm (developed by our collaborators from Harvard University) to automatically segment the mitotic spindle in DIC movies of C. elegans embryos, determine the position of centrosomes, quantify the morphology and motions of the spindle, and track these features over time. - Statistical analysis using Matlab. *Scientific communication - Posters for scientific congress - Two scientific articles (one published and one in progress) *Supervision of trainees Master 1 student (1 month) et 2nd year technician student (4 months) Responsabilities: - Manage lab's stock - Update lab's database for plasmid, primers, strains Show less
