My master’s thesis was focused on differentiating various lines of human pluripotent and induced pluripotent stem cell towards pancreatic islet-like cells. Differentiated cells were transplanted under the kidney capsule of immunocompromised mice for the further studies. The purpose of my thesis work was to optimize differentiation protocol and techniques to generate functionally more mature islet-like cells, and generate islet-like cells and transplants for single-cell RNA sequencing. The thesis work and other projects, that I assisted in, included cell culturing, FACS, RT-qPCR, laboratory mice research (handling, blood sampling, injecting, surgery), immunohistology (fixing, embedding, sectioning, staining), fluorescence microscopy and preparing cell samples for single-cell RNA sequencing.
