  Researched various methods to discover how to change the specificity of a riboswitch from Guanine to Hypoxanthine by in vitro selection. Focused on the biological roles and activities of a family of enzymes called ADARs given they catalyze the reaction that converts adenosine to inosine within double-stranded DNA. Used methods such as Reverse Transcriptase Polymerase Chain Reaction (RT-PCR), RNA splicing, fluorescence, UV shadowing, RNA purification, and in vitro selection throughout the duration of the research experiment. Analyzed the selected RNA on both native and denaturing gels. Applied the concept of ligand-binding to troubleshoot and problem solve new ways to successfully complete the project.   Focused on polyacrylamide gel electrophoresis to separate specific DNA and RNA fragments within the tested substances. Conducted multiple rounds of RNA Polymerase Chain Reactions to isolate and amplify the desired RNA. Utilized Microsoft Excel and Word to analyze and compile the data into a cohesive report. Gained information from the Excel spreadsheets and lab report to determine the strength of certain DNA fragments to resist radiation poisoning. This supports the mission of AFRRI to protect the military members against radiation threats in the present and future. Developed a strong understanding of the importance of precision in science. Created multiple dilutions of varying concentrations to produce the desired results through the gel. Solidified proper lab procedures when reading the gels to include allowing proper prep time for the gels to develop before proceeding with dark room analysis. Show less
