My PhD project is a collaboration between the laboratory of Neural Stem Cells and Neurogenesis with Dr Juan Manuel Encinas and the laboratory of Neuronal Excitability with Dr Jan Tonnesen. During this project, I have set a model of epileptic hippocampal organotypic slices (hOTCs) to study how seizures induce reactive neurogenesis, subsequently disrupting synapses and circuitry. The organotypic slice model allows for fast and efficient pharmacological and genetic modifications of signaling pathways. It also allows for high resolution imaging (by 2-photon microscopy) coupled to electrophysiological recordings (from calcium imaging to microelectrode arrays). Thus, we investigate alterations in newborn neurons and their impact on the synapse and functional connectivity. Show less

As part of my thesis project, I made an international stay of 4 months at the Laboratory of Neurodevelopment and Adult Neurogenesis in Epilepsy supervised by Prof. Jenny Hsieh at UTSA in San Antonio, Texas. By injecting RVDREADDs (hm4Di) with AAV.Syn.GCamp and RVGCamp in hOTCs, I was able to assess neural network general activity and newborn neuron specific activity by calcium imaging, which was supported by subsequent MEA recordings. I had the opportunity of doing an 8 month internship in the Neurovascular laboratory in the Vall d'Hebron Research Institute in Barcelona, Spain, with the supervision of the Dra. Anna Rosell. During this period, I studied the influence of Diesel Exhaust Particles (DEP) as a major air pollutant in brain, where I made the analysis of the particles, assessed their effect in angiogenesis in EPCs in vitro and studied their acute effect in vivo. During the period of internship in the Laboratory of Biochemistry and Molecular Biology of the University of the Basque Country (UPV/EHU) I developed a proteomics study focused on the CRM1 protein. Here I carried out the information-compilation and design of a targeted proteomics experiment with the supervision of the Dra. Miren Josu Omaetxebarria and the Dr. Javier Beaskoetxea Lejarzegi.
