Project Title: Regulation of placental autophagy in the context of malaria in pregnancy. Objective: Poor pregnancy outcomes that characterize malaria in pregnancy are often associated with dysregulation of placental homeostasis due to inflammation triggered in response to Plasmodium spp. infections. Mechanisms associated with homeostasis maintenance such as autophagy might be affected, having consequences to the healthy development of pregnancy. Therefore, the project aimed to analyze autophagy-related proteins such as LC3 and SQSTM1/p62 in placentas from pregnant women and mice with malaria during pregnancy to evaluate if inflammation triggered in response to Plasmodium spp. modulates placental autophagy during pregnancy. Project Title: A novel approach to identify the specific antibody characteristics important for protection from malaria in pregnant women. Objective: Little is known about immunity to malaria in pregnancy and the importance of VAR2CSA immunity in protecting pregnant women from malaria in low transmission settings. Thus, the project focused on the antibody responses to VAR2CSA and non-pregnancy-related P. falciparum antigens in a population of Brazilian pregnant women living in malaria endemic regions. Project Title: Impact of Plasmodium infection on the expression of bradykinin-receptor 2 on trophoblasts. Objective: Intrauterine growth restriction is a hallmark of malaria in pregnancy due to placental insufficiency promoted by abnormal vascularity and dysregulation of vasomodulatory factors. Molecular mechanisms that govern these outcomes are still unclear. Herein, we evaluated the expression of the vasodilator bradykinin receptor 2 on the surface of trophoblasts from pregnant mice infected with P. berghei to establish a link between placental vasomodulation and malaria associated poor pregnancy outcomes. Project Title: Establishment of a placental malaria mouse model using transgenic P. berghei expressing the full length of the VAR2CSA protein. Objective: Preclinical models to study immunity acquisition to VAR2CSA are scarce due to the absence of known orthologs in the murine parasites that can be studied. Herein, we assessed the pathogenesis of murine malaria in pregnancy induced by a transgenic P. berghei expressing the full length of VAR2CSA. Supervision: Dr. Luciana Vieira de Moraes, Dr. Carlos Penha-Gonçalves. Show less
