      Student internship carried out in agreement at the Biogem Genetic Research Institute in Ariano Irpino, under the supervision of the Headmaster Prof. Ambrosino C. The 16-hour training experience had the following aims: Part I: Execution of a PCR and procedure for preparing an agarose gel and displaying electrophoretic run. Next, a cell culture setup of HEK293 cells was created on a Petri dish. Part II: Performing a minipreparation of plasmid DNA for the extraction, on a small scale, of plasmid DNA. Performing a digestion reaction with restriction enzymes. Performing a lentiviral transfection protocol on cells cultured with vesicular stomatitis virus G protein (VSV-G), as well as components of the packaging construct (Gag - Pol - Rev) and of GFP protein. Last but not least, a Burker Chamber was set up in the cell chamber to count the cells in culture and estimate their number and percentage viable. Show less
