Research Project: Designed and tested antisense oligonucleotides (ASOs) targeting a locus relevant to Alzheimer’s Disease as a potential gene therapy approach for the treatment of AD. This project provided a novel therapeutic strategy for AD treatment which has now been patented and a manuscript submitted for publication purposes. Skills:- Improved project planning skills by independent planning of each step of my PhD project and supervising the projects of MSc students- Enhanced oral communication skills by presenting at multiple symposiums and conferences- Enhanced written communication skills demonstrated by writing a 50,000-word scientific thesis, as well as various reviews and a research paper which has been submitted for publication - Data analysis and statistical analysis skills demonstrated by use of SPSS, GraphPad Prism 7.0, and ImageJ as well as the analysis of massive data set throughout PhD- Organisation skills evidenced by the organised records of data and lab books which were kept throughout PhDLaboratory skills and techniques:- RNA analysis - RT-PCR, RT-qPCR, Gel electrophoresis- Protein analysis – DC assays, SDS-PAGE, Western blotting, Immunocytochemistry and confocal microscopy- Tissue culture – Cultured SH-SY5Y, HEK293T, Hela and Neuro2A cells, cell cryopreservation, cell differentiation, cell transfection- ASO design – Use of SpliceFinder and various other bioinformatic tools- CRISPR-CAS9 – Guide design and cloning for both SA-CAS9 and SP-CAS9 guides
