Thesis title - Insights into copper tolerance in Staphylococcus aureus The main objective of this thesis is to study the growth profiles and copper tolerance of four S. aureus strains with different genomic background at different copper concentrations and to perform the biochemical characterization of the multicopper oxidase (MCO) enzyme from S. aureus. During this internship, I acquire laboratory experience in expression and purification of heterologous proteins, which include techniques such as chromatography, SDS-PAGE, PAGE, E. coli transformation, and in the biochemistry and enzymatic characterization of enzymes, in this case the multicopper oxidase, and growth profiles of S. aureus. Internship - Molecular Microbiology of Bacterial Pathogens Laboratory (FCT-UNL)

In this voluntary internship I studied the Staphylococcus aureus bacteria and developed laboratory experience in Molecular Biology and Microbiology. The laboratory procedures performed and learned during the internship consisted in preparations of culture media, molecular cloning, DNA extraction and purification, conventional and pulsed field gel electrophoresis (PFGE), PCR and biochemistry assays (S. aureus cell wall hydrolysis assays). Internship - Plant Molecular Ecophysiology Laboratory

This volunteer internship was made in the are of knowledge of plant molecular biology where I adcquired laboratory experience in preparation of solutions (stock and working solutions), RNA extraction from grapes berries, spectrophotometric quantification of RNA, nucleic acids electrophoresis. I also learned to work with liquid nitrogen. In this internship I collaborated with a project related to marine ecology, namely in biological monitoring in the “Ribeiras de Cascais” project, where the treatment of samples of benthic macroinvertebrates was carried out to assess the quality of the water in the streams.
