Aim: Elucidate potential therapeutic effects of OT-1/mb-IL15 against solid tumor in lymphoreplete mice. Role: Performed transduction of murine CD8+ OT-1 cells with mb-IL15 molecular construct. Performed adoptive cell transfers of CD8+ OT-1 expressing mb-IL15 to C57BL/6J mice. Designed flow cytometry panels using 8-10 colors. Utilized BD LSRII and LSRFortessa flow cytometers. Handled mice and performed IV and IP injections. Performed tail nick and cardiac puncture for blood collection. Immunostaining of splenocytes and whole blood. Used FlowJo, GraphPad Prism, and BD FACSDIVA software Show less

Aim: Engineer TCR/CAR T cells with IL-15 to generate synergistic effects between host immune cells and TCR/CAR T cells against solid tumors and enhance expansion of TCR/CAR T cells in lymphoreplete mice. Role: Designed and developed mbIL-15 molecular constructs using In-fusion cloning technique and SnapGene software. Subcloned molecular construct to retroviral expression vector. Performed transfections for retrovirus production. Executed Isolation, activation and transduction of murine CD8a T cells. Utilized flow cytometry BD FACSCalibur and FlowJo to measure transduction efficiency of CAR and mb-IL15 molecular constructs. Executed intracellular staining to detect IL-15 signaling. Performed flow cytometry-based killing assays. Designed and executed experiments to block IL15 signaling using mABs Show less

Aim: Execute cell-based assays using proprietary CAR T technology to assess their anti-tumor immunity and safety switch. Role: Performed antibody-dependent cellular cytotoxicity (ADCC) assays, complementdependent cytotoxicity (CDC) assays and killing assays. Analyze cell-based assays using iQue flow cytometer and FlowJo software. Isolation of PBMCs from leukopaks.
